"Mnie" is used with dopełniacz (kogo? czego?), and biernik (kogo? co?), while "mi" is used with celownik (komu? czemu?):

Also, when you want to emphasize that it is "me" at the beginning or end of the sentence, you can use the longer form "mnie", instead of "mi".

I find it a bit confusing, why some sentences use mnie instead of mi. The other sentence, like "could you please tell me" , used "mi" instead of "mnie" although both mean "me" in English. Can you explain more about this?

I don't see the Polish phrase for "Do you have a medial prescription, sir?" in the Lesson Materials. It would be helpful to have that written out too. The individual words aren't in the vocabulary list either.
